Cause Breast Cancer?

Science (July 21, 2010) — Women who report greater use of cleaning products may be at higher breast cancer risk than those who say they use them sparingly. Researchers writing in BioMed Central’s open access journal Environmental Health asked more than 1500 women about their cleaning product usage and found that women who reported using more air fresheners and products for mold and mildew control had a higher incidence of breast cancer.


Julia Brody, from the Silent Spring Institute, USA, worked with a team of researchers to carry out telephone interviews with 787 women diagnosed with breast cancer and 721 comparison women. She said, “Women who reported the highest combined cleaning product use had a doubled risk of breast cancer compared to those with the lowest reported use. Use of air fresheners and products for mold and mildew control were associated with increased risk. To our knowledge, this is the first published report on cleaning product use and risk of breast cancer.”

The researchers questioned women on product use, beliefs about breast cancer causes, and established and suspected risk factors. They found that cleaning products, air fresheners, and insect repellents were associated with breast cancer, but little association was observed with overall pesticide use. Women with breast cancer who believed that chemicals and pollutants contribute ‘a lot’ to the risk of developing the condition were more likely to report high product usage.

Speaking about this potential bias to the study, Brody said, “When women are diagnosed with breast cancer, they often think about what happened in the past that might have contributed to the disease. As a result, it may be that women with breast cancer more accurately recall their past product use or even over-estimate it. Or, it could also be that experience with breast cancer influences beliefs about its causes. For example, women diagnosed with breast cancer are less likely to believe heredity contributes ‘a lot’, because most are the first in their family to get the disease.”

In order to avoid possible recall bias, the researchers recommend further study of cleaning products and breast cancer using prospective self-reports and measurements in environmental and biological media.

Red Wine Vs. White?

Both Equal Offenders In Breast

-Cancer Risk

Science (Mar. 10, 2009) — The largest study of its kind to evaluate the effect of red versus white wine on breast-cancer risk concludes that both are equal offenders when it comes to increasing breast-cancer risk. The results of the study, led by researchers at Fred Hutchinson Cancer Research Center, were published in the March issue of Cancer Epidemiology, Biomarkers and Prevention.


“We were interested in teasing out red wine’s effects on breast-cancer risk. There is reason to suspect that red wine might have beneficial effects based on previous studies of heart disease and prostate cancer,” said lead author Polly Newcomb, Ph.D., M.P.H., head of the Cancer Prevention Program in the Public Health Sciences Division at the Hutchinson Center. “The general evidence is that alcohol consumption overall increases breast-cancer risk, but the other studies made us wonder whether red wine might in fact have some positive value.”

Instead, Newcomb and colleagues found no compelling reason to choose Chianti over Chardonnay.

“We found no difference between red or white wine in relation to breast-cancer risk. Neither appears to have any benefits,” Newcomb said. “If a woman drinks, she should do so in moderation – no more than one drink a day. And if a woman chooses red wine, she should do so because she likes the taste, not because she thinks it may reduce her risk of breast cancer,” she said.

The researchers found that women who consumed 14 or more drinks per week, regardless of the type (wine, liquor or beer), faced a 24 percent increase in breast cancer compared with non-drinkers.

For the study, the researchers interviewed 6,327 women with breast cancer and 7,558 age-matched controls about their frequency of alcohol consumption (red wine, white wine, liquor and beer) and other breast-cancer risk factors, such as age at first pregnancy, family history of breast cancer and postmenopausal hormone use. The study participants, ages 20 to 69, were from Wisconsin, Massachusetts and New Hampshire. The frequency of alcohol consumption was similar in both groups, and equal proportions of women in both groups reported consuming red and white wine.

The National Cancer Institute, a branch of the National Institutes of Health, funded this research, which also involved investigators from Group Health Cooperative, Seattle; the University of Wisconsin; H. Lee Moffitt Cancer Center & Research Institute; and Dartmouth Medical School.

Women Who Choose Boiled Coffee

Run Lower Risk of Breast Cancer,

Swedish Study Finds

Science (June 16, 2010) — Women who drink Scandinavian boiled coffee, which chemically resembles French press and Turkish/Greek coffee, more than four times a day run a lower risk of developing breast cancer than women who drink coffee less than once a day. This is shown by Lena Nilsson and her associates at Umeå University in an article in the journal Cancer Causes & Control.


A major difference between boiled and filtered coffee is that the boiled version contains up to 80 times as much coffee-specific fatty acids. These fatty acids have previously been shown in animal experiments to inhibit the growth of cancer.

By comparing filtered coffee and boiled coffee in the Västerbotten Intervention Project (64,603 participants), researchers at Umeå University have been able to show for the first time that various brewing techniques can lead to different risk patterns for cancer. For total cancer, prostate cancer, colorectal cancer, and many other less common forms of cancer, there was no correlation.

Among women who drank boiled coffee more than four times a day there was a lowered risk of breast cancer compared with women who drank coffee less than once a day. Among women who drank filtered coffee there was an increased risk for early breast cancer (under 49 years old) and a decreased risk for late breast cancer (over 55 years old). Boiled-coffee drinkers, but not filtered-coffee drinkers, also had an increased risk of pancreatic cancer and lung cancer among men.

The study, recently published in the scientific journal Cancer Causes and Control, is the first in the world to compare the consumption of coffee prepared with two brewing techniques in regard to cancer.

Science (July 13, 2008) — A gene that is overexpressed in 20 percent of breast cancers increases the number of cancer stem cells, the cells that fuel a tumor’s growth and spread, according to a new study from the University of Michigan Comprehensive Cancer Center.


The gene, HER2, causes cancer stem cells to multiply and spread, explaining why HER2 has been linked to a more aggressive type of breast cancer and to metastatic disease, in which the cancer has spread beyond the breast, the researchers say.

Further, the drug Herceptin, which is used to treat HER2-positive breast cancer, was found to target and destroy the cancer stem cells. “This work suggests that the reason drugs that target HER2, such as Herceptin and Lapatanib, are so effective in breast cancer is that they target the cancer stem cell population. This finding provides further evidence for the cancer stem cell hypothesis,” says study author Max S. Wicha, M.D., Distinguished Professor of Oncology and director of the U-M Comprehensive Cancer Center.

The cancer stem cell hypothesis says that tumors originate in a small number of cells, called cancer stem cells, and that these cells are responsible for fueling a tumor’s growth. These cells represent fewer than 5 percent of the cells in a tumor. Wicha’s lab was part of the team that first identified stem cells in human breast cancer in 2003.

In the current study, researchers found that breast cancer cells overexpressing the HER2 gene had four to five times more cancer stem cells, compared to HER2-negative cancers. In addition, the HER2-positive cells caused the cancer stem cells to invade surrounding tissue, suggesting that HER2 is driving the invasiveness and spread of cancer.

The researchers then looked at the drug Herceptin, which is used to treat HER2-positive breast cancer. They found Herceptin reduced the number of cancer stem cells in the HER2-positive breast cancer cell lines by 80 percent, dropping it to the same levels seen in HER2-negative cell lines.

When HER2 was not overexpressed in the cell cultures, the researchers found, the cancer stem cell population did not increase. Nor did Herceptin have any effect on the HER2-negative cells, which is consistent with how Herceptin is used in the clinic.

“We are now studying what pathways are activated by HER2 overexpression. Our hope is that we could develop inhibitors of these pathways that might be effective in targeting cancer stem cells in women whose tumors do not overexpress HER2 or those who are resistant to Herceptin,” says study author Hasan Korkaya, Ph.D., a U-M research fellow in internal medicine.

Breast cancer statistics: 184,450 Americans will be diagnosed with breast cancer this year and 40,930 will die from the disease, according to the American Cancer Society. About 20 percent of breast cancers are considered HER2-positive.

Study Finds

Science (May 5, 2010) — A compound derived from broccoli could help prevent or treat breast cancer by targeting cancer stem cells — the small number of cells that fuel a tumor’s growth — according to a new study from researchers at the University of Michigan Comprehensive Cancer Center.


The study tested sulforaphane, a component of broccoli and broccoli sprouts, in both mice and cell cultures. Researchers found sulforaphane targeted and killed the cancer stem cells and prevented new tumors from growing.

“Sulforaphane has been studied previously for its effects on cancer, but this study shows that its benefit is in inhibiting the breast cancer stem cells. This new insight suggests the potential of sulforaphane or broccoli extract to prevent or treat cancer by targeting the critical cancer stem cells,” says study author Duxin Sun, Ph.D., associate professor of pharmaceutical sciences at the U-M College of Pharmacy and a researcher with the U-M Comprehensive Cancer Center.

Results of the study appear in the May 1 issue of Clinical Cancer Research.

Current chemotherapies do not work against cancer stem cells, which is why cancer recurs and spreads. Researchers believe that eliminating the cancer stem cells is key to controlling cancer.

In the current study, researchers took mice with breast cancer and injected varying concentrations of sulforaphane from the broccoli extract. Researchers then used several established methods to assess the number of cancer stem cells in the tumors. These measures showed a marked decrease in the cancer stem cell population after treatment with sulforaphane, with little effect on the normal cells. Further, cancer cells from mice treated with sulforaphane were unable to generate new tumors. The researchers then tested sulforaphane on human breast cancer cell cultures in the lab, finding similar decreases in the cancer stem cells.

“This research suggests a potential new treatment that could be combined with other compounds to target breast cancer stem cells. Developing treatments that effectively target the cancer stem cell population is essential for improving outcomes,” says study author Max S. Wicha, M.D., Distinguished Professor of Oncology and director of the U-M Comprehensive Cancer Center.

The concentrations of sulforaphane used in the study were higher than what can be achieved by eating broccoli or broccoli sprouts. Prior research suggests the concentrations needed to impact cancer can be absorbed by the body from the broccoli extract, but side effects are not known. While the extract is available in capsule form as a supplement, concentrations are unregulated and will vary.

This work has not been tested in patients, and patients are not encouraged to add sulforaphane supplements to their diet at this time.

Researchers are currently developing a method to extract and preserve sulforaphane and will be developing a clinical trial to test sulforaphane as a prevention and treatment for breast cancer. No clinical trial is currently available.
